If you put the video card in properly, it should snap in. You'll know when it's actually in cause you can feel it, and it makes a distinct sound. And then the screw should be the one from those long pieces of metal you had to remove in order to have the ports of the video card stick out of your tower. It doesn't matter if you put the card in before or after the standoffs, although I'd do it after.
Like snap when you put it into the motherboard slot?
Also, my case didn't have screws to take out the back slots. You just had to pull out the metal, then there's this locking mechanism inside that I'm supposed to use (optional), but I can't seem to get it to work. I'll try it again, though.
